dc.description.abstractCAAX amino-terminal protease familien (heretter omtalt som CAAX-proteiner), består av antatt membranbundne metallproteaser. Dette er proteiner som finnes i både eukaryote- og prokaryote celler, men deres funksjon i prokaryote er fortsatt ukjent. I eukaryote celler spalter disse proteinene av -AAX tripeptidet hos prenylerte proteiner som inneholder den C-terminale sekvensen CAAX. Streptococcus pneumoniae ble brukt som modellorganisme i dette arbeidet for å studere CAAX-proteiner i bakterier. I 2010 kom det ut en studie som tyder på at CAAX-proteiner er involvert i sekresjonen av virulensfaktorer i Staphylococcus aureus. S. pneumoniae, i likhet med S. aureus, sekreterer de fleste proteinene sine over den cytoplasmatiske membranen via Sec sekresjonsmaskineriet. Dette er et sekresjonsmaskineri som sekreterer ufoldede proteiner med et hydrofobt signalpeptid (også kalt ledersekvens). Dette signalpeptidet kløyves av det ufoldede proteinet etter fullført translokasjon. I S. pneumoniae er det identifisert to typer signalpeptider som bruker dette sekresjonsmaskineriet: de med normale hydrofobe signalsekvenser og de som i tillegg har et såkalt YSIRK-motiv. Hovedmålsetningen for dette arbeidet var å karakterisere CAAX-proteinene i S. pneumoniae, og spesifikt studere hvordan disse proteinene og ledersekvenser påvirker sekresjonen av viktige virulensfaktorer. I dette arbeidet ble CAAX-genene identifisert som ikke-essensielle gener i S. pneumoniae. Det ble ikke observert noen karakteristisk fenotype med hensyn på vekst, antibiotikasensitivitet eller cellemorfologi i mutanter hvor alle 10 genene som koder for CAAX-proteiner var deletert. CAAX-proteinene er membranproteiner som lokaliserer hovedsakelig til septum av cellen, selv om det er noe variasjon mellom de ulike CAAX-proteinene med hensyn på lokalisering. Disse forskjellene tyder på at proteinene kan ha ulike funksjoner i cellen. Sekresjon av virulensfaktorer med ulike ledersekvenser (PspA og PspC) ble studert i villtypeceller og mutanter hvor CAAX-genene var fjernet. Det ble ikke funnet noe som tyder på at CAAX-proteiner er involvert i sekresjon av virulensfaktorer i S. pneumoniae. Basert på resultatene i denne oppgaven, er det derfor ikke mulig å konkludere med hvilke funksjoner CAAX proteinene har i S. pneumoniae. Under arbeidet med PspA og PspC, ble det derimot oppdaget at ledersekvensen er avgjørende for hvordan kolinbindende proteiner sekreteres i S. pneumoniae. Mens normale ledersekvenser fører til sekresjon ved cellepolene, gir ledersekvenser med såkalt YSIRK-motiv sekresjon i celledelingssonen i midten av cellen. En slik funksjon for YSIRK-ledersekvenser har ikke tidligere blitt rapportert i S. pneumoniaenb_NO
dc.description.abstractThe CAAX-aminoterminal protease family (from now on referred to as CAAX-proteins), consists of putative membrane-bound metalloproteases. These proteins are found in both eukaryotic and prokaryotic cells, but their function in prokaryotes is still unknown. In eukaryotic cells, these proteins cleave the -AAX tripeptide in prenylated proteins containing the C-terminal sequence CAAX. Streptococcus pneumoniae was used as a model organism in this work to study CAAX-proteins in bacteria. A study in 2010 suggested that CAAX-proteins are involved in the secretion of virulence factors in Staphylococcus aureus. S. pneumoniae, like S. aureus, secretes most of its proteins across the cytoplasmic membrane via Sec secretion machinery. This is a secretion machinery that secretes unfolded proteins with a hydrophobic signal peptide (also called leader sequence). The signal peptide is cleaved off the unfolded protein after translocation. In S. pneumoniae two types of signal peptides are identified: those with normal hydrophobic signal sequence and those which additionally have a so-called YSIRK-motif. The main goal of this study was to characterize the CAAX-proteins in S. pneumoniae, and specifically study how these proteins and leader sequences influence the secretion of important virulence factors. In this study, CAAX-genes were identified as non-essential genes in S. pneumoniae. No characteristic phenotype was observed with respect to growth, antibiotic sensitivity or cell morphology in mutants where all 10 genes encoding CAAX-proteins were deleted. The CAAX-proteins are membrane proteins that mainly localize to the septum of the cell, although there is some variation among the different CAAX-proteins with regard to localization. These differences indicate that the proteins may have different functions in the cell. Secretion of virulence factors with different leader sequences (PspA and PspC) was studied in wild-type cells and mutants where the CAAX-genes were removed. There was no evidence that CAAX-proteins are involved in secretion of virulence factors in S. pneumoniae. Based on the results of this study, it is therefore not possible to conclude which functions CAAX-proteins have in S. pneumoniae. When working with PspA and PspC it was discovered that the leader sequence is crucial for secretion of choline binding proteins of S. pneumoniae. While normal leader sequences lead to secretion at the cell poles, leader sequences with so-called YSIRK-motif give secretion in the cell division site in the center of the cell. Such function of YSIRK leader sequences has not previously been reported in S. pneumoniae.nb_NO
