dc.description.abstract | Cancer is a group of deadly diseases worldwide, and in 2013 more than 30 000 new cancer cases were reported in Norway. If cancer is detected before dissemination, there are good prospects of being successfully treated with current technology. However, it is the spread of cancer that causes most deaths related to cancer. Therefore it is of interest to find preventive measures, which can slow the dissemination, thus increasing the success rate of treatment and reduce the recurrence of cancer.
Production of microalgae started in the early 1900s, and the use of microalgae in biotechnology has long been focusing on nutrients produced by the algae. Lately, the focus has also been directed to the use of algae in pharmaceuticals. Thus it has been reported that the extract of Chlorella sorokiniana inhibits migration and induces apoptosis in cancer cells (Hep G2). In this thesis it was invastigated whether the extract from C. sorokiniana may reduce cell viability, inhibit migration through Matrigel and fibrin gel, and induce apoptosis in cancer cells. In the experiments, two extractants, ethanol (96%) and water (dH2O), were used for examining the extraction procedure that affected the cell lines the most. The algae extracts were named according to extraction solvent used, CSEt (ethanol) and CSH (dH2O)
In the MTT viability experiments the cell lines WI-38 (normal lung fibroblasts), WI-38 VA-13 (SV40-transformed) and Hep G2 (hepatocellular carcinoma) were treated with algea extracts. The greatest decrease in cell viability was observed in cell lines treated with CSEt. It was also observed a decrease in cell viability for the same cell lines treated with CSH. However, the effect of these extracts was more moderate compared to CSEt. Based on migration assay tests there was no indication of algea extracts inhibiting cell invation in either fibrin gel or Matrigel of the Hep G2 cell line.
In the immunoblot experiments was examined whether algae extracts induced apoptosis in Hep G2 cell line. No activated caspase-3 was identified in the experiments, and if the decrease in cell viability observed in the MTT viability experiments were do to apoptosis can not be confirmed. Further work is needed to identify what caused the cell death observed in MTT viability experiments. | nb_NO |

legemidler.
Det
har
blant
annet
blitt
rapportert
at
ekstrakt
fra
Chlorella
sorokiniana
inhiberer
migrering
og
induserer
apoptose
i
kreftceller.
I
denne
masteroppgaven
skal
det
undersøkes
om
ekstrakt
fra
C.
sorokiniana
kan
redusere
viabiliteten,
inhibere
migrasjon
gjennom
Matrigel
og
fibringel,
samt
indusere
apoptose
i
kreftceller
(Hep
G2).
I
forsøkene
ble
det
brukt
to
ekstraksjonsmidler,
96%
etanol
(EtOH)
og
vann
(dH2O),
for
å
undersøke
hvilken
ekstraksjonsmetode
som
påvirket
kreftcellene
mest.
Algeekstraktene
ble
navngitt
i
henhold
til
ekstraksjonsmiddel
som
ble
brukt,
CSEt
(etanol)
og
CSH
(dH2O)
.
I
MTT-‐viabilitets-‐forsøkene
ble
cellelinjene
WI-‐38
(normale
lungefibroblaster),
WI-‐38
VA-‐13
(SV40-‐transformert)
og
Hep
G2
(hepatocellulært
karsinom)
behandlet
med
algeekstraktene.
Det
ble
observert
størst
nedgang
i
celleviabilitet
i
cellelinjene
behandlet
med
CSEt.
Det
ble
også
observert
en
nedgang
i
celleviabilitet
for
de
samme
cellelinjene
behandlet
med
CSH,
men
effekten
av
disse
ekstraktene
var
mer
moderat
enn
effekten
av
CSEt.
Ut
i
fra
migrasjonsassayforsøkene
så
det
ikke
ut
til
at
algeekstraktene
inhiberte
migrasjon
av
Hep
G2-‐cellelinjen
verken
gjennom
fibringel
eller
Matrigel.
I
immunoblotforsøkene
skulle
det
undersøkes
om
algeekstraktene
induserte
apoptose
i
Hep
G2-‐cellelinjen.
Det
ble
ikke
identifisert
aktivert
caspase-‐3
i
noen
av
forsøkene,
og
om
nedgangen
i
celleviabilitet
som
ble
observert
i
MTT-‐viabilitets-‐forsøkene
var
apoptose-‐relatert,
kan
ikke
bekreftes.
Videre
arbeid
er
nødvendig
for
å
kunne
identifisere
hva
som
forårsaket
celledøden
i
