dc.description.abstractAndoSan™ er et medisinsk soppekstrakt der hovedkomponenten er den champignonlignede soppen Agaricus blazei Murill (AbM). Preparatet inneholder også to andre sopparter, Hericium erinaceum og Grifola frondosa. De siste årene har det vært mye oppmerksomhet rundt ekstraktet da det antydes å ha positiv effekt på en rekke sykdomstilstander. Hvilke komponenter og virkningsmekanismer som er årsaken til dette er fortsatt uklart, men det antas at karbohydrater og proteiner spiller en rolle. Målet med denne oppgaven har vært å undersøke proteiner i AndoSan™ og eventuelt påvise hva slags type proteiner dette var. Presipitering ble gjort for å oppkonsentrere proteindelen av ekstraktet. Videre ble det gjort analyseringer av proteinkonsentrasjon og proteinprofiler, sammen med identifisering av proteiner med LC-MS analyse og Mascot databasesøk. Det ble også undersøkt om proteinene ble fordøyd i et in vitro modellsystem med humane gastrointestinale enzymer. Proteinprofil og identifisering av peptider ble utført på de fordøyde prøvene. Måling av proteinkonsentrasjon og analysering av proteinprofiler viste at det fantes proteiner i ekstraktet, samtidig som det tydet på at konsentrasjonen var lav. Mange av proteinbåndene ble først påvist etter farging med 0,1 % sølvnitrat. Resultatene fra LC-MS analyse og Mascot databasesøk viste at flere proteiner ble identifisert, blant annet aktin, histon H4 og endo-xylanase. Disse proteinene ble identifisert i ulike proteinbånd separert på Bio-Rad geler (12 % og any kD) og hadde MW på ca. 97, 30 og 14,4 kDa. Ved in vitro fordøyelse med humane fordøyelses enzymer viste SDS-PAGE resultatene at mange av proteinene i ekstraktet lot seg nedbryte. Samtidig ble det påvist fire peptidsekvenser etter fordøyelse med human gastrisk juice (HGJ). Disse hadde en lengde på 11-15 aminosyrer og tilhørte proteinet aktin. Resultatene fra analysene som er blitt gjort viser at det finnes proteiner i AndoSan™. Det er imidlertid flere usikkerhetsmomenter knyttet til resultatene og for å kunne konkludere med tanke på proteininnhold og type proteiner i AndoSan™ bør videre analysering utføres. Det er også usikkert om de identifiserte proteinene og peptidene har betydning i forbindelse med den foreslåtte virkningen AndoSan™ har på immunforsvaret og kreft. Videre arbeid kan for eksempel undersøke om peptidsekvensene identifisert etter in vitro fordøyelsen, har noen effekt på kreftceller. AndoSan™ is a mushroom extract that is used as an alternative remedy. The main component in AndoSan™ is Agaricus blazei Murill, but the extract also contain two other mushrooms, Hericium erinaceum and Grifola frondosa. In the last years, the extract has been subjected to much attention since it has shown to have positive effect on various diseases. Which components and mechanisms that cause this is rather unclear, but it is assumed that carbohydrates and proteins is involved. The aim of this thesis has been to investigate proteins in AndoSan™ and possibly detect what kind of proteins this is. Precipitation was done to concentrate the protein part of the extract. Further, analysis was done to determine the protein concentration and protein profiles, together with identification of proteins with LC-MS analysis and Mascot database search. The digestion of proteins was also investigated with use of an in vitro model system with human gastrointestinal enzymes. Protein profiles and identification of peptides were done on the digested fractions. Measuring of the protein concentration and analysis of protein profiles showed that there were proteins in the extract, but the concentration was low. Many of the protein bands were detected after silver staining. Further analysis with LC-MS and Mascot database search showed that several proteins were identified, including actin, histone H4 and endo-xylanase. These proteins were identified from different protein bands separated on Bio-Rad gels (12 % and any kD) and had MW about 97, 30 and 14,4 kDa. The SDS-PAGE results from the in vitro digestion showed that many proteins were digested. Four peptides were also identified after digestion with human gastric juice (HGJ). These were 11-15 amino acids long, and belonged to the actin protein. The results from the different analyses show that there are proteins in AndoSan™. It is however, several uncertainties related to the results and to conclude further analysis has to be done. It is also uncertain whether the identified proteins and peptides have any effect associated with the proposed effect AndoSan™ has shown on the immune system and cancer. Further work can be done to investigate if the peptides identified after the in vitro digestion has any effect on cancer cells.nb_NO
