Smør og margariner er fettrike produkter som for mange er en naturlig del av det daglige
kostholdet. En forutsetning for å kunne gjøre en ernæringsmessig vurdering av en matvare er å
ha kjennskap til produktenes sammensetning. Matens fettsyresammensetting har i de senere år
fått mye oppmerksomhet på grunn av helsemessige virkninger av fettsyrer. Spesielt har det blitt
påpekt at inntaket av langkjedede omega-3 fettsyrer bør økes, og at mengden mettede fettsyrer
bør begrenses. Sammenliknet med smør konsumeres det mye mer av margarinprodukter. De
inneholder omega-3 fettsyren α-linolensyre som mennesker til en viss grad kan omdanne til
eikosapentaensyre (EPA) og dokosaheksaensyre (DHA). I tillegg til at fettsyresammensetningen
har en helsemessig betydning, vil måten fettsyrene frigjøres og hvor raskt de frigjøres også kunne
ha betydning.
I denne oppgaven har formålet vært å undersøke graden av hydrolyse av lipider i vann- i
olje baserte produkter med forskjellig innhold og sammensetning av fett. En to trinns in vitro
fordøyelsesmodell med human magesaft (HGJ) og tynntarmssaft (HDJ) ble brukt for å simulere
fordøyelse av de kommersielle produktene smør, margarin og lettmargarin. Første trinn med
magesaft i 60 min og deretter trinn 2 med tarmsaft i 60 og 120 minutter. Etter fordøyelse ble
fettklassene triacylglyserol (TAG) og frie fettsyrer (FFA) isolert ved bruk av fast fase ekstraksjon
(solid phase extraction, SPE). Fettsyresammensetningen i de to fraksjonene ble kvalitativt og
kvantitativt analysert ved gasskromatografi massespektrometri (GC-MS).
Triacylglyserolene i margarinprøvene hadde et høyt innhold av umettede fettsyrer (65 %),
og sammensetningen reflekterte klart at produktene inneholder en god del rapsolje, som
inneholder mye av fettsyren 18:1n-9. Smør hadde et høyt innhold av mettede fettsyrer (67 %), der
16:0 dominerte. Smørprøvene inneholdt i tillegg en del korte fettsyrer som 4:0; 6:0 og 8:0 som i
stor grad stammer fra den mikrobielle fordøyelsen i vomma hos drøvtyggere.
I trinnet som simulerte magefordøyelsen ble det observert svært liten dannelse av frie
fettsyrer i alle prøvene. Videre hydrolyse med tarmsaft i 60 min resulterte i en høy frigjøring av
fettsyrer fra TAG. Mindre hydrolyse av TAG ble observert fra 60 min til 120 minutters inkubering
med tarmsaft. I følge analyser i blod fordøyes 98 % av fettet i kosten. Nedbrytingen av TAGs i
dette prosjektet stanset ved ca 35 - 40 %. Årsaken kan være at FFA ikke ble fjernet som i en in
vivo fordøyelse, og dette etablerer en likevekt i reaksjonen som hindrer at all TAG hydrolyseres.
Arbeidet med denne oppgaven har vist at bruk av human fordøyelsessaft i et in vitro
fordøyelsessystem kan gi nyttig informasjon om hvordan fordøyelsen av fett i ulike matprodukter
skjer. Resultatene i dette prosjektet la imidlertid ikke grunnlag for å konkludere med at det var
forskjeller i fordøyelsen av de ulike produktene. Det gjenstår mye med hensyn til optimalisering
av reaksjonsbetingelsene. I tillegg bør prøvetakingstidspunktene kanskje være oftere. Ekstraksjon
og analyse av lipidene som dannes ved inkubering kan også forbedres. Abstract
Butter and margarine are products which are rich in fat and which for many people form a regular
part of the daily diet. In order to perform a nutritional evaluation of a foodstuff, knowledge of the
products composition is required. In recent years, foods fatty acid composition has been the
center of a lot of focus. This is primarily due to the health-related effects of fatty acids. In
particular, it has been highlighted that intake of long chained omega-3 fatty acids should be
increased while that the amount of saturated fatty acids in the diet should be limited. In
comparison with butter, consumption of margarine products is much higher. These margarine
products contain the omega-3 fatty acid α-linolenic acid which humans are capable of converting
into eicosapentaenoic (EPA) and docosahexaenoic (DHA). In addition to the health related effects
of the fatty acid composition of food, the manner in which the fatty acids are released and how
quickly they are released, may also be significant.
The aim of the current study was to investigate the degree of lipid hydrolysis in water-oil
based products, with varying fat content and fat composition. A two step in vitro human digestive
model, with human gastric juice (HGJ) and human duodenal juice (HDJ), was employed to
stimulate digestion of the following commercial products; butter, margarine and light margarine.
The first step comprised of gastric juice for 60 minutes while the next step involved duodenal
juice for 60 and 120 minutes respectively. Following digestion, the classes of fat triacylglycerol
(TAG) and the free fatty acid (FFA) were isolated using solid phase extraction (SPE). The two
sections of the fatty acid composition were qualitatively and quantitatively analyzed with the aid
of gas chromatography mass spectrometry (GC-MS).
The triacylglycerols in the margarine samples had a high content of unsaturated fatty acids
(65%). Furthermore their composition clearly reflected the products high content of rapeseed oil,
which contains a lot of the fatty acid 18:1n-9. Butter contained a lot of saturated fatty acids (67%),
the majority of which were 16:0. Butter also contained a certain degree of short fatty acids such
as 4:0; 6:0 and 8:0, which predominantly arise from microbial digestion in the paunch of
ruminating mammals. In the gastric digestion stimulating step, very little free fatty acid formation
was observed in all the samples. Further hydrolysis with duodenal juice in 60 minutes resulted in
a high release of fatty acids from TAG. Reduced hydrolysis of TAG was observed from 60 to 120
minutes incubation with duodenal juice. According to analysis of blood, 98% of dietary fat is
digested. However decomposition of TAGs in this project ceased at approximately 35-40%. The
reason behind this may be that FFA was not removed, as would have occurred in an in vivo
digestion. This may have established an equilibrium in the reaction, which subsequently
prevented complete TAG hydrolysis.
The work carried out in connection with this study, has shown that the use of human
digestive juice in an in vitro can provide useful information concerning how digestion of fat, in
various food products, occurs. The results in this project cannot however provide a reliable basis
for concluding that there is variations in the digestion of the various products. With respect to
optimization of the conditions surrounding the various reactions in this study, a certain amount of
work still remains. In addition should perhaps sampling frequency occur more often. Extraction
and analysis of the lipids formed during the incubation may also be improved.
