Repetitive regioner i Lactobacillus reuteri celle - og mucusbindende protein A (CmbA) - studier av variasjon og betydning for adhesjon til intestinale celler

Sammendrag
Hovedmål i dette arbeidet var å studere om den hypervariable repeterte regionen i den C-terminale delen av CmbA har betydning for adhesjon til intestinale epitelceller, og om forskjellige vekstfaser og mediekomponenter kan påvirke uttrykket av cmbA-genet vedrørende disse repetitive regionene.
I dette arbeidet ble det brukt to syntetisk produserte varianter av cmbA-genet, tilstede i plasmidene pUC57-cmbA3 og pUC57-cmbA4. Det lyktes å transformere cmbA3-varianten inn i L. reuteri 6475cmbA- i en konstruksjon med vektor pSIP411. Stammen ble sammen med pSIP-cmbA1 testet for adhesjon til Caco-2. Siden det ikke lyktes å finne positive transformanter av cmbA4 i hverken Lc. lactis og E. coli PK401 kan det tenkes at genet, som er 3.4 kb, kanskje er for stort, men også at genet har egenskaper som gjør det vanskelig å introdusere i ulike stammer. Siden antall repetitive regioner er det som skiller genene kan det hende at akkurat denne ekstra delen i cmbA4 forårsaker vanskelighetene. DSM20016 og L. reuteri 6475 ble benyttet i vekstforsøk for å analysere de repetitive regionene i cmbA. Galle og mucin ble brukt som mediekomponenter for å se om disse påvirket resultatet. DSM20016 og 6475 tilsatt galle vokste betydelig saktere enn de andre bakteriekulturene, midlertidig økte veksten ved tilsats av MOPS-buffer. Det kan antas at MOPS hindrer stor hemming i bakteriekulturene med galle. Fra PCR-produkt var det ingen signifikante forskjeller i de repetitive mønstrene, men i DSM20016 kan det tenkes at mucin kanskje induserer større variasjon og eventuell forskjell til binding tatt i betraktning at mucin hadde høyere utbytte i båndene på slutten av vekstforsøket enn kontrollen. I 6475 har kontrollen antydninger til flere bånd enn 6475 med mucin og galle i tillegg til et høyere utbytte i hvert bånd. Dette kan tyde på at mucin eller galle hemmer evnen til større variasjon i repeterende regioner i 6475. The overall aim in this study was to see if the hypervariable repetitive region in

It was speculated that the si
ze of the gene (3,4kb) might be too big for introduction in the
cells. In addition, the gene has properties that
make it difficult to introduce in different strains.
Since the number of repetitive regions is what separates the genes it might be this partic
ular
extra
part
in
cmbA4
which causes difficulties.
DSM20016 and
L. reuteri
6475 were u
sed in the growth experiment to
analyze the repetitive
regions. Bile and mucin
was used as media component to
see if these affected the result. In
this experiment the s
trains with bile grew significantly slower tha
n the other bacterial
cultures, though the growth increased with addition of MOPS
-
buffer. MOPS might prevent
inhibiting in the cultures with bile.
DSM20016 and 6475 with mucin grew fastest.
There were
no signif
icant differences in the repetitive regions in the patterns from PCR. However, in
DSM20016 it might be that mucin induces greater variation and difference in binding,
considering that mucin had higher yield in the end of the growth experiment than the cont
rol
(DSM20016 without mucin). 6475 had
more bands than 6475 with bile and mucin, this could
mean that mucin or bile inhibits the
ability to greater variation
in
the repetitive regions.
