The physiological and genetic basis for yield formation and maturation behavior in faba bean (Vicia faba L.) under Norwegian climate

Faba bean (Vicia faba L.) is a cool-season grain legume cultivated as a spring crop at higher latitudes such as the Nordic region, while in more southern regions and subtropical zones, it is grown as a winter or spring crop. The importance of faba bean within human diets and animal nutrition is significant as it is a notable source of protein, starch, minerals, vitamins, and dietary fibers. Faba bean's pivotal role within agriculture extends to its capacity for biological nitrogen fixation through a symbiosis with Rhizobium bacteria, which makes it a resource-efficient and suitable crop to be included in the rotation. Despite the inherent benefits associated with faba bean, its cultivation has remained restricted in Norway mainly because of low yield stability, persistent challenges linked to pests and diseases, and limitations in breeding programs.
Consequently, enhancing faba bean's appeal as a crop to agronomists and farmers entails the introduction of novel cultivars, characterized by high and stable yield, and resistance to diseases. Progress in the selection and breeding of faba bean requires efficient and costeffective methods to evaluate large numbers of genotypes. Traditional data capturing by manual methods is labor-intensive, destructive, and prone to human errors. Remote sensing platforms equipped with different sensors have been widely used in other crops and are among the most influential technologies for smart farming and precision agriculture as well as efficient and non-destructive techniques for high throughput phenotyping in breeding.
Objectives of this study were to assess the diversity presented within spring-type cultivars, identify stable faba bean cultivars well-suited to the high-latitude, cool, and humid climate of Norway for yield, thousand kernel weight (TKW), and growing degree days (GDD) to reach BBCH 89 (maturation). Another objective was to assess the potential utility of RGB and multispectral sensors to estimate plant height, predict SPAD values (an indicator of chlorophyll content) and yield. The third objective was to assess the impact of cultivar selection, environments, and the application of fungicide on the development of chocolate spot (CS) disease (Botrytis fabae) in faba bean and utilize multispectral data to devise a streamlined monitoring protocol for CS.
Comprehensive field experiments including a selection of 38 spring-type cultivars sourced from European breeding companies were conducted at three locations in Norway in 2021, 2022 and 2023 growing seasons. Notably, this research marks the first extensive assessment of such a substantial number of cultivars across diverse locations within Norway. The effects of cultivar, environment and their interaction were assessed on fifteen phenological, physiological and agronomic traits of faba bean. The additive main effects and multiplicative interaction (AMMI) model and coefficient of variation (CV) were used to assess the stability of cultivars for yield, TKW, and GDD to BBCH 89. Multispectral and RGB aerial images were collected on a weekly basis, spanning from the sowing to the harvest time, using Phantom 4 multispectral camera and DJI Phantom 4 respectively. Spectral reflectance values of different bands (red, green, blue, near infrared, and red edge), digital surface model (DSM), and digital terrain model (DTM) were calculated at the plot level. Different spectral indices were calculated based on the spectral values of different bands for prediction and classification of faba bean traits and DSM generated from RGB images were used to estimate plant height. Moreover, the machine learning algorithms Support Vector regression and Random Forest with two feature selection methods, Pearson correlation coefficient (PCC), and Sequential Forward Feature Selection (SFS), were used to predict yield and SPAD value. This study also characterized the spectral signatures of faba bean plants affected by CS and assessed the viability of employing a Support Vector Machine (SVM) for the classification of CS severity.
The results of this study showed that faba bean yield was highly correlated with TKW and GDD to reach BBCH 89 (maturation). Stability analysis identified cultivars Birgit, Stella, Bobas, and Macho as the most stable high-yielding cultivars achieving a mean yield of 6.1- 6.4 tons ha-1. Bobas, Macho, Stella, and Yukon had the most stable TKW (612-699 g) and Bobas, Capri, Trumpet, and Vertigo were the most stable to reach BBCH 89 (1257°C days). These stable cultivars can be utilized in breeding programs with the purpose of breeding for high and stable yield and TKW, as well as for adaptation to the Norwegian climate. The correlation between manual and UAV-based plant height measurements revealed a strong agreement with a correlation coefficient (R2) of 0.97. The best prediction of SPAD value was achieved at BBCH 50 (flower bud present) with an R2 of 0.38 and root mean square error (RMSE) of 1.14. For yield prediction, BBCH 60 (first flower open) was identified as the optimal stage, using spectral indices with an R2 of 0.83 and RMSE of 0.53 tons ha-1. This developmental stage presents an opportunity to implement targeted management practices to enhance yield.
Moreover, significant variability was observed in cultivars' susceptibility to CS, with Bolivia exhibiting the highest level of resistance and Louhi, Sampo, Vire, Merlin, Mistral, and GL Sunrise being the most susceptible. Fungicide application significantly reduced CS severity and improved yield components. Analysis of canopy spectral signatures revealed the near infrared (NIR) and red edge bands, along with enhanced vegetation index (EVI) and soil adjusted vegetation index (SAVI), as the most useful spectral bands and indices to estimate CS severity because they had a strong negative correlation with CS severity ranging from -0.51 to -0.71. In addition, EVI enabled early detection of CS and abiotic stresses in the field. SVM accurately classified CS severity into four classes (resistant, moderately resistant, moderately susceptible, and susceptible) based on spectral data with higher accuracy after the onset of disease compared to later in the season (accuracy 0.75- 0.90).
In conclusion, this comprehensive field study sheds light on the intricate dynamics of faba bean cultivation, highlighting the significant impact of both genetic factors and environmental variables on key agronomic traits. The identification of stable, high-yielding cultivars like Birgit, Stella, Bobas, and Macho presents promising prospects for the advancement of faba bean breeding programs in Norway. Furthermore, the integration of UAV technology equipped with RGB and multispectral sensors and machine learning algorithms emerges as a valuable tool for efficient phenotyping and CS monitoring. Åkerbønne (Vicia faba L.) er en kjernebelgvekst tilpasset kjølige dyrkingsforhold. Åkebønne dyrkes som en vårsådd vekst på høyere breddegrader som i Norden, mens den i mer sørlige regioner og subtropiske soner dyrkes vår- eller høstsådd. Betydningen av åkerbønne i kostholdet og i fôr til husdyr har blitt stadig mer fremtredende ettersom den utmerker seg som en betydelig kilde til protein, stivelse, mineraler, vitaminer og kostfiber. Åkerbønnens sentrale rolle i jordbruket begrunnes også med dens evne til biologisk nitrogenfiksering gjennom en symbiose med Rhizobium-bakterier, noe som gjør den til en ressurseffektiv og viktig vekst å inkludere i vekstskiftet. Til tross for fordelene assosiert med åkerbønne, har dyrkingen vært begrenset i Norge, hovedsakelig på grunn av lav avlingsstabilitet, vedvarende utfordringer knyttet til skadedyr og sykdommer, samt begrenset avlsarbeid for å tilpasse sorter til norske dyrkingsmiljø.
Følgelig innebærer det å forbedre åkerbønnens anvendelse for agronomer og bønder innføringen av nye sorter som er kjennetegnet av høy og stabil avling og motstand mot sykdommer. Fremgang i utvalg og avl av åkerbønne krever effektive og kostnadseffektive metoder for å evaluere et stort antall genotyper. De tradisjonelle, manuelle metodene er ofte arbeidsintensive, destruktive og utsatt for menneskelige feil. Fjernmålingsplattformer utstyrt med ulike sensorer har vært en mye brukt og innflytelsesrik teknologi for smart jordbruk og presisjonslandbruk, og kan være effektive og ikke-destruktive teknikker for datainnsamling.
Ett av målene for dette studiet var å analysere diversitet i et utvalg av vår-sådde åkerbønnesorter, og å identifisere stabile sorter av åkerbønne tilpasset nordlige dyrkingsområder med et kjølig og fuktig klima. Mål nummer to var å undersøke mulighetene for å benytte RGB og multispektrale sensorer for å estimere plantehøyde, og for å prediktere SPAD verdier (en indikator for plantenes klorofyll innhold) og avling. Det tredje målet var å analysere effekter av sortsvalg, dyrkingsmiljø og behandling med fungicid på utvikling av sykdommen sjokoladeflekk (CS) (Botrytis fabae) i åkerbønne, og å utnytte multispektrale data for å etablere og protokollere en effektiv overvåkningsmetode for CS.
Det er gjennomført omfattende feltforsøk med et utvalg av 38 sorter av vår-åkerbønne innhentet fra Europeiske planteforedlere, og som ble anlagt på tre lokaliteter i Norge i sesongene 2021, 2022 og 2023. Dette er første gang det blir gjennomført så omfattende utprøving av åkerbønnesorter i Norge. Effektene av sorter, dyrkingsmiljø ble vurdert på femten fenologiske, fysiologiske og agronomiske egenskaper av åkerbønne. AMMI modellen (Additive Main effects and Multiplicative Interaction) og variasjonskoeffisienter (CV) ble brukt for å analysere sortenes stabilitet i avling, TKW og varmesum ved modning (BBCH 89). Multispektrale og RGB bilder ble samlet inn ukentlig, fra såing til høsting, ved hjelp av Phantom 4 multispektral kamera og DJI Phantom 4. Spektral refleksjonsverdier for ulike bånd (rød, grønn, blå, nærinfrarøde og rødkant), digital overflate modell (DSM) og digitalt terreng modell (DTM) ble beregnet på plott-nivå. Forskjellige spektrale indekser ble beregnet basert på spektralverdiene for ulike bånd for å prediktere og klassifisere egenskaper av åkerbønne. DSM fra RGB bilder var brukt for å estimere plantehøyde. Maskinlæringsalgoritmene Support Vector og Random Forest med to ulike seleksjonsmetoder, Pearsons korrelasjons koeffisient (PCC) og Sequential Forward Feature Selection (SFS), ble brukt for å prediktere avling og SPAD verdier. Studien har også karakterisert de spektrale signaturerne til åkerbønneplanter som er angrepet av CS og det er brukt Support Vector Machine (SVM) for å klassifisere angrepsgraden av CS.
Resultatet av denne studien viste at avling var sterkt korrelert med TKW og nødvendige graddager for å nå BBCH 89 (modning). Stabilitetsanalyse identifiserte sortene Birgit, Stella, Bobas og Macho som de mest stabile og yterike sortene med et gjennomsnittlig avlingsutbytte på 6.1-6.4 tonn ha-1. Bobas, Macho, Stella og Yukon hadde den mest stabile TKW (612.2-699 g) og Bobas, Capri, Trumpet og Vertigo var de mest stabile for å nå BBCH 89 (1257°C dager). Disse stabile sortene kan brukes i avlsprogrammer med formålet om høy og stabil avling, og TKW, samt å være tilpasset det norske klimaet. Korrelasjonen mellom manuell og UAV-baserte plantehøydemålinger viste en sterk enighet med en korrelasjonskoeffisient (R2) på 0.97. Den beste forutsigelsen av SPAD-verdi ble oppnådd på BBCH 50 (blomsterknopp til stede) med en R2 på 0.38 og RMSE på 1.14. For avlingsprediksjon ble BBCH 60 (første blomst åpen) identifisert som det optimale stadiet, ved bruk av spektrale indekser med en R2 på 0.83 og RMSE på 0.53 tonn ha-1. Dette utviklingsstadiet presenterer en mulighet for å implementere målrettede forvaltningspraksiser for å forbedre avlingen.
I tillegg ble det observert betydelig variabilitet i sortenes mottakelighet for CS, med Bolivia som viste det høyeste nivået av motstand og Louhi, Sampo, Vire, Merlin, Mistral og GL Sunrise som de mest mottakelige. Behandling med fungicid reduserte sykdomsutvikling av CS og forbedret avlingskomponenter. Analyse av spektrale signaturer avslørte at nærinfrarøde (NIR) og rødkantbåndene, sammen med forbedret vegetasjonsindeks (EVI) og jordjustert vegetasjonsindeks (SAVI), var mest følsomme for CS-infeksjon og de hadde en sterk negativ korrelasjon med CS-alvorlighetsgraden som varierte fra -0.51 til -0.71. I tillegg muliggjorde EVI tidlig deteksjon av CS og abiotiske stress i feltet. SVM klassifiserte nøyaktig CS-alvorlighetsgraden i fire klasser (motstandsdyktig, moderat motstandsdyktig, moderat mottakelig og mottakelig) basert på spektrale data med høyere nøyaktighet etter sykdommens utbrudd sammenlignet med senere i sesongen (nøyaktighet 0.75- 0.9).
Avslutningsvis kaster denne omfattende feltstudien lys over de intrikate dynamikkene i dyrkingen av åkerbønne, og fremhever den betydelige innvirkningen av både genetiske faktorer og miljøvariabler på viktig agronomiske egenskaper. Identifiseringen av stabile, høyavlingssorter som Birgit, Stella, Bobas og Macho presenterer lovende utsikter for avansementet av åkerbønneavlsprogrammer i Norge. Videre fremstår integreringen av UAV-teknologi utstyrt med avansert bildebehandling og maskinlæringsalgoritmer som et verdifullt verktøy for effektiv fenotyping og CS.
