dc.description.abstractBackground: An increase in the incidence of pathogenic Vibrio bacteria is expected in Nordic waters as a result of global warming. Vibrio-bacteria can cause infections that in some cases require treatment with antibiotics. Knowledge of antimicrobial-resistance among these bacteria is therefore important. The spread of antimicrobial resistance (AMR) is relatively well understood in clinical contexts, but the role of the aquatic environment in the spread of AMR is still largely unknown. The aquatic environment can be a place for the development and spread of AMR. It is a comprehensive process to understand and thus requires scientific knowledge across disciplines for the fight against the development and spread of AMR in different environments. Determination of AMR among bacterial strains is necessary to monitor changes in bacterial susceptibility to antibiotics. Purpose: This study aims to investigate the occurrences of V. parahaemolyticus and the characterization of virulence and the AMR in these bacteria extracted from mussels, seaweed, and seawater collected from Norway's eastern and western coasts. Materials and methods: A total of 6 samples of 72 mussels (Mytilus edulis), and 6 samples of 600 grams of bladderwrack seaweed (Fucus vesiculosus) were examined by the Most probable number method (MPN) for enumeration of E. coli, and Vibrio bacteria according to NMKL method 156 1997. 6 samples of 3000 ml seawater were examined for the presence of thermotolerant coliform bacteria and Vibrio spp. The bacterial isolates were identified by Gram and oxidase testing, API 20E and MALDI-TOF MS. In addition, the bacterial isolates were tested for antibiotic sensitivity and determination of hemolysis activity. Results: In the Vibrio method, 113 colonies were isolated from mussels, seaweed, and seawater. A total of 38 colonies were identified using MALDI-TOF. Ten strains were identified as V. parahaemolyticus. Bacterial isolates were Gram-negative and oxidase positive. Furthermore, no β-hemolysis was detected in all isolated V. parahaemolyticus. All bacterial isolates were sensitive to the 12 different antibiotics tested for in this study. Conclusion: The main findings in this study show that V. parahaemolyticus isolated from mussels, seaweed, and seawater is generally sensitive to antibiotics. This study further shows that the incidence of V. parahaemolyticus in seafood is low and there is an absence of hemolysis. Given that climate change is likely to increase the incidence of the human pathogenic Vibrio, more frequent monitoring of the incidence, resistance, and virulence of Vibrio bacteria is necessary to prepare for future public health challenges.en_US

Vibrio-bakterier kan gi infeksjoner som i noen tilfeller krever behandling med antibiotika. Kunnskapen om antimikrobiell resistent blant disse bakteriene er derfor viktig. Spredningene av antimikrobiell resistens er godt forstått i klinisk sammenheng, men vannmiljøets rolle i spredningen av AMR er fortsatt ukjent. Vannmiljøet kan være et sted for utvikling og spredning av AMR. Det er en omfattende prosess å forstå og krever dermed vitenskapelig kunnskap på tvers av fagfelt for kampen mot utvikling og spredning av AMR i ulike miljøer. Bestemmelse av AMR blant bakteriestammer er nødvendig for å overvåke endringer i bakterienes følsomhet for antibiotika. Hensikt: Denne studien har som mål å undersøke forekomster av V. parahaemolyticus og karakterisering av virulens og AMR hos disse i prøver fra blåskjell, blæretang og sjøvann som er samlet fra øst- og vestkysten av Norge. Materialer og metoder: Totalt 6 prøver av 72 blåskjell (Mytilus edulis), 6 prøver av 600 gram blæretang (Fucus vesiculosus) ble undersøkt i en fem ganger tre-rørs mest sannsynlige tall metode for telling av E. coli, samt Vibrio bakterier i henhold til NMKL metode 156 1997. 6 prøver av 3000 ml sjøvann ble undersøkt med tanke på forekomsten av termotolerante koliforme bakterier og Vibrio spp. Bakterieisolatene ble identifisert ved hjelp av Gram- og oksidase testing, samt API 20E og MALDI-TOF MS. I tillegg ble bakterieisolatene testet for antibiotika følsomhet og bestemmelse av hemolyseaktivitet. Resultater: I Vibrio-metoden ble det isolert 113 kolonier isolert fra blåskjell, blæretang og sjøvann. Totalt 38 ble identifisert ved hjelp av MALDI-TOF. Ti stammer ble identifisert som V. parahaemolyticus. Bakterieisolatene var Gram-negative og oksidasepositive. Videre ble ingen β-hemolyse påvist hos noen av de isolerte V. parahaemolyticus. Alle bakterieisolater var følsomme for de 12 forskjellige antibiotika som det ble testet for i denne studien. Konklusjon: Hovedfunnene i denne studien viser at V. parahaemolyticus isolert fra blåskjell, blæretang og sjøvann gjennomgående er følsomme for antibiotika. Denne studien viser videre at forekomsten av V. parahaemolyticus i sjømat er lav og det er fravær av hemolyse hos isolatene. Gitt at klimaendringer sannsynligvis vil gi økt forekomst av det humanpatogene Vibrio, er hyppigere overvåking av forekomst, resistens og virulens hos Vibrio-bakterier nødvendig for å forberede seg på fremtidige folkehelseutfordringer.en_US
