dc.description.abstractHensikten med denne oppgaven var å undersøke populasjonen av hvite blodceller i råmelk fra ku og i blod fra deres respektive kalver. I tillegg ble en eventuell sammenheng mellom hvite blodceller i råmelk og i blod hos kalv undersøkt. Forsøket ble gjennomført ved Senter for Husdyrforsøk (SHF, NMBU, Ås) og studien er basert på råmelksprøver fra ni kyr av rasen Norsk rødt fe (NRF) og blodprøver fra deres ti kalver (ett sett tvillinger). Prøvene ble tatt like etter fødsel før tildeling av råmelk, og deretter 48 timer etter fødsel. Prøvene ble analysert ved bruk av flow-cytometri. Det er godt etablert at råmelk (kolostrum) inneholder store mengder antistoffer og at disse er avgjørende for den nyfødte kalvens motstandskraft mot sykdom. Årsaken til at kalven har behov for en overføring av maternale antistoffer er at morkaka (placenta) hindrer overføring av antistoffer under drektigheten. Det har i senere tid blitt undersøkt om mordyrets hvite blodceller (maternale leukocytter) blir overført til kalvens sirkulasjonssystem via kolostrum, og om disse har en betydning for kalvens immunforsvar. For å undersøke endringene i leukocytter i råmelk og i blod hos kalv ble en Wilcoxon signed- rank-test gjennomført. For å undersøke en eventuell sammenheng mellom leukocytter i råmelk og i blod hos kalv ble Spearman-korrelasjon brukt. I råmelk ble det observert signifikant økning i andelen gd T-celler (P=0,039) og monocytter (P=0,023), samt en tendens til økning i CD3-celler (P=0,098) og CD8-celler (P=0,055). I kalvenes blod var det signifikante økning i andel gd T-celler (P=0,039) og WC1-celler (P=0,027), samt en signifikant reduksjon for granulocytter (P=0,012). I tillegg ble det observert en tendens til reduksjon andel i NK-celler (P=0,051). Det er også vist en korrelasjon mellom CD8-celler og WC1-celler i råmelk på dag 1 og i kalveblod på dag 2, henholdsvis P=0,001 og P=0,078. Disse funnene indikerer en sammenheng mellom type og andel leukocytter i råmelk fra mor og innholdet i kalvens blod. Dette støtter teorien om en overføring av maternale leukocytter via råmelk til kalvens blod, som vist gjennom tidligere studier. I fremtidig forskning bør en slik overføring undersøkes ytterligere, og i hvilken grad dette er av positiv betydning for kalvens immunforsvar og motstandskraft tidlig i livet.nb_NO
dc.description.abstractThe aim of this study was to investigate the population of leukocytes in colostrum of the mothers and also in the blood of their neonatal calves. In addition, a possible relationship between leukocytes in colostrum and in neonatal calf blood was examined. The research was conducted at the Center for livestock production (SHF, NMBU, Ås, Norway), and was based on colostrum samples from nine Norwegian Red cows and blood samples of their ten calves (one set of twins). The first sampling was performed just after birth before colostrum feeding, and the second sampling at approximately 48 hours after birth. The analysis of the samples was performed by flow cytometry. Colostrum contains a large amount of maternal antibodies that are essential for the neonatal calf and its resistance against illness. The reason for the need of maternal antibodies transferred from colostrum is the impermeability of the bovine placenta during gestation. In recent research it has been studied that the substantial number of maternal leukocytes in colostrum may transfer to the calf ́s circulation and contribute to the calf ́s immune system. To investigate the changes of leukocyte distribution in colostrum and calf blood, a Wilcoxon signed-rank-test was performed. A possible relationship between leukocytes in colostrum and in calf blood was examined, and the Spearman correlation was used. Presented results show changes in leukocyte population in both colostrum and calf ́s blood from parturition until two days postpartum. In colostrum it was observed significant increase in portion of gd T-cells (P=0,039) and monocytes (P=0,023), and a tendency for increase of CD3 (P=0,098) and CD8 cells (P=0,055). In calf blood there was found significant increase in the proportion of gd T- cells (P=0,039) and WC1 cells (P=0,027), a significant decrease of granulocytes (P=0,012) as well as a tendency for reduction of NK-cells (P=0,051). Further, we found correlation between proportion of CD8 cells and WC1 cells in colostrum at day 0 and in calf blood at day 2, P=0,001 and P=0,078 respectively. These findings indicate that there might be a relation between type and portion on leukocytes in colostrum and in the blood of the neonatal calf ́s blood. These findings support the theory whether maternal leukocytes transfer to the neonatal calf by colostrum as shown by earlier studies. In future reaserch this should investigated further, especially to elucidate the impact these leukocytes have on the calf ́s immunity system.nb_NO
