dc.description.abstractNæringsrik melk fra storfe har en sentral rolle i det daglige kostholdet til de fleste nordmenn. God melkekvalitet er viktig for å opprettholde melkeforbruket, og i tillegg er det viktig for bonden som får betaling etter kvalitetsgraden av melk. Melkekvaliteten bedømmes etter flere forskjellige kvalitetsparametere, og for forbrukeren er det spesielt melkens lukt og smak som har stor betydning. Melken blir analysert for innholdet av frie fettsyrer, da økt lipolyse av fett i melk resulterer i dårligere smak på melk. Det er flere faktorer som kan påvirke innholdet av frie fettsyrer i melk, blant annet fôringen av melkekyrne. Av hovednæringsstoffene i melk er fett den komponenten som kan påvirkes i størst mulig grad gjennom fôringen. Palmitinsyre er kvantitativt viktigst av fettsyrene i melkefett, og i tillegg har den ofte en sentral rolle i kraftfôr til høytytende kyr. For å opprettholde melkeytelsen og fettinnholdet i melk i tidlig laktasjon, blir det vanligvis tilsatt ekstra fôrfett i kraftfôret. Det mest brukte tilskuddsfett i dag er palmeoljebasert, noe som inneholder en vesentlig andel palmitinsyre. Hovedformålet med masteroppgaven var derfor å undersøke sammenhengen mellom innholdet av palmitinsyre og frie fettsyrer i melk, og i tillegg om en eventuell sammenheng kan relateres til fôringen av kyrne. Denne masteroppgaven består av en teoretisk del og en eksperimentell del. Den teoretiske delen omhandler betydningen av fett i rasjonen til melkekyr. Litteraturdelen omhandler først oppbygging av fett, fett i fôr og fordøyelsen av hovednæringsstoffene i vom og tarm med spesielt vekt på fettfordøyelsen. Melk inneholder 3-5% fett og fettsyrene i melk består av både mettede- og umettede fettsyrer, der mettede fettsyrer dominerer og utgjør ca. 64% av totalt fett i melk. De tre kvantitativt viktigste fettsyrene i melk er palmitinsyre, stearinsyre og oljesyre. Sammensetningen av melk og melkefett gjennomgås i litteraturdelen, og i tillegg forhold ved fôret som påvirker melkefett. Fettsyrene som finnes i melk stammer fra de novo syntesen i juret og fettsyrer tilført juret via blodet. Melkefett er inkorporert i fettkuler som er omgitt av en kompleks membran som fungerer som beskyttelse mot lipolyse av fettet i melk som kan bidra til smaksfeil. Dette omtales i litteraturdelen, etterfulgt av en gjennomgang av melkekvalitet og regelverket omkring kvalitetsparametere, inkludert melkens innhold av frie fettsyrer. Fôringen av melkekyrne og flere andre faktorer som kan påvirke innholdet av frie fettsyrer i melk, vil gjennomgås til slutt i litteraturdelen. Litteraturdelen støtter en positiv sammenheng mellom innholdet av palmitinsyre og frie fettsyrer i melk, men sikker dokumentasjon mangler. Den eksperimentelle delen omhandler en statistisk analyse for å undersøke om melkens innhold av frie fettsyrer har sammenheng med innholdet av mettede fettsyrer, monoumettede fettsyrer, polyumettede fettsyrer og umettede fettsyrer i melk, og i tillegg til enkeltfettsyrene palmitinsyre, stearinsyre og oljesyre i melk. Det ble også undersøkt om innholdet av frie fettsyrer har sammenheng med innholdet av fett, laktose, protein og urea i melk. Den statistiske analysen ble gjort på analyseresultatene av tankmelkprøver for årene 2015 og 2016 fra utvalgte besetninger som leverer melk til TINE Råvare. I tillegg ble det testet om det er sammenheng mellom melkens innhold av frie fettsyrer og data fra Kukontrollen over kraftfôrmengde og melkeavdrått. Resultatet av den statistiske analysen for sammenhengen mellom innhold av palmitinsyre og frie fettsyrer i melk viste en positiv signifikant sammenheng (P<0,0001), noe som bekreftet hypotesen i masteroppgaven. Det ble funnet negativ sammenheng mellom melkens innhold av mettet fett og frie fettsyrer (P<0,0001), til tross for at melkens innhold av palmitinsyre økte ved økt innhold av frie fettsyrer. Mengde fettsyrer syntetisert de novo var dermed negativt korrelert til innholdet av frie fettsyrer i melk. Det kan ha sammenheng med fôring av kyrne, og det ble funnet positiv sammenheng mellom innhold av frie fettsyrer i melk og forbruk av kg kraftfôr pr. 100 kg EKM (P<0,0001). En reduksjon av de novo syntetiserte fettsyrer vil vanligvis føre til lavere fettinnhold i melk, men i denne undersøkelsen økte fettinnholdet ved økt innhold av frie fettsyrer i melk (P<0,0001). Melkens innhold av fett kan være påvirket av innholdet av palmitinsyre, da denne fettsyren kan bidra til økt fettinnhold i melk. Innholdet av umettede fettsyrer viste positiv sammenheng med innhold av frie fettsyrer i melk (P<0,0001). Dette gjaldt også monoumettede fettsyrer (P<0,0001). Økningen i innholdet av umettet fett er sannsynligvis sterkt påvirket av oljesyre som også viste positiv sammenheng med innholdet av frie fettsyrer i melk (P<0,0001), da den er veldig viktig kvantitativt sett. Derimot ble det påvist en reduksjon av polyumettede fettsyrer (P=0,0002) ved økt innhold av frie fettsyrer i melken, noe som indikerer at melkens innhold av polyumettede fettsyrer kan forebygge økt innhold av frie fettsyrer i melk.nb_NO
dc.description.abstractNutritious milk from cattle has a central role in the daily diet of many Norwegians. Good milk quality is important to maintain milk consumption, and in addition, it is important for the milk farmer who receive payment according to the quality of milk. The milk quality is determined by several different quality parameters, and for the consumer, the smell and taste of the milk are of particular importance. The content of free fatty acids is analysed in milk, as increased lipolysis in milk results in the appearance of rancid flavours. Several factors can affect the content of free fatty acids in milk, including the feeding of the cows. Of the main nutrients in milk, the one most easily to affect by feeding is fat. Palmitic acid is quantitatively the most important of the fatty acids in milk and in addition, it often plays a central role in concentrates given to high-yielding cows. In order to maintain milk yield and fat content in milk in early lactation, supplementation of diets with fat is usual. The most commonly used dietary fat supplement today is palm oil based, which contain a substantial amount of palmitic acid. The main purpose of this master thesis was therefore to study the relationship between the content of palmitic acid and free fatty acids in milk, and in addition, evaluate whether any correlation can relate to the feeding of the cows. This master's thesis consists of a theoretical part and an experimental part. The theoretical part deals with the significance of fat in the feed of dairy cows. The literature section reviews the structure of fat, fat in feed and the digestion of the main nutrients through the four-compartment stomach and the intestinal tract, with particular emphasis on fat digestion. Milk contains 3-5% fat and the fatty acids in milk consist of both saturated- and unsaturated fatty acids. Saturated fatty acids dominate in milk and account for about 64% of total fat. The three quantitatively most important fatty acids in milk are palmitic acid, stearic acid and oleic acid. The literature section reviews the composition of milk and milk fat, as well as aspects of the feed affecting milk fat. The fatty acids found in milk origins from the de novo fatty acid synthesis in the udder and fatty acids applied to the udder from the bloodstream. Milk fat is present as globules covered by a complex membrane, which acts as protection against lipolysis of milk fat that can result in rancid flavour in milk. The literature section reviews this, as well as milk quality and the regulations concerning quality parameters including milk’s free fatty acid content. The feeding of the cows and several other factors, which can affect the content of free fatty acids in milk, are reviewed at the end of the literature section. The literature section supports a positive correlation between the content of palmitic acid and free fatty acids in milk, but reliable documentation is scarce. The experimental section deals with a statistical analysis to determine if the content of free fatty acids in milk are correlated to the content of saturated fatty acids, monounsaturated fatty acids, polyunsaturated fatty acids and the total of unsaturated fatty acids in milk, in addition to the single fatty acids palmitic acid, stearic acid and oleic acid in milk. The correlation between the milk’s content of free fatty acids and the content of fat, lactose, protein and urea in milk were also determined. The statistical analysis were performed on analysis results of milk samples from selected milk farms, which were delivering milk to TINE Råvare in 2015 and 2016. In addition, the correlation between the content of free fatty acids in milk and data of the amount on concentrates and milk yield from Kukontrollen were tested. The result of the statistical analysis of the relationship between the content of palmitic acid and free fatty acids in milk showed a positive significant relationship (P<0,0001), which confirmed the hypothesis of this master’s thesis. A negative relationship between the content of saturated fatty acids and free fatty acids were found (P<0,0001), despite the fact that the milk’s palmitic acid content increased as the content of free fatty acids increased. Thus, the amount of fatty acids synthesized de novo were negative correlated to the content of free fatty acids in milk. This may be related to the feeding of the cows, and a positive correlation were found between the content of free fatty acids in milk and the consumption of kg concentrates per 100 kg ECM (P<0,0001). A reduction of fatty acids synthesized de novo usually results in lower fat content in milk, but in this study, the fat content increased as the content of free fatty acids increased in milk (P<0,0001). The content of palmitic acid in milk may have affected the fat content in milk, as this fatty acid can influence an increase of fat content in milk. A positive correlation between the content of unsaturated fatty acids and free fatty acids were found (P<0,0001). This also applied to monounsaturated fatty acids (P<0,0001). The increase in the content of unsaturated fat is most likely by the influence of the content of oleic acid, as it is very important quantitatively. The results showed a positive correlation between the content of free fatty acid and oleic acid in milk (P<0,0001). On the other hand, the results also showed a decrease of the content polyunsaturated fatty acids as the content of free fatty acids increased in milk (P=0,0002). This indicates that the milk’s content of polyunsaturated fatty acids can prevent an increase of free fatty acids in milk.nb_NO
