dc.description.abstractSammendrag Hovedfunksjonen til den bakterielle celleveggen er å motstå det intracellulære trykket og å opprettholde morfologien til bakteriene. Skjelettet i celleveggen består av peptidoglykan. Strukturen til peptidoglykan er kompleks, og selv om det har vært studert i mange år har man enda ikke en full forståelse av dets tredimensjonale struktur og hvordan dette makromolekylet syntetiseres. Celledeling og celleveggsyntese hos bakterier er nøye regulerte og koordinerte prosesser hvor mange komponenter inngår. Streptococcus pneumoniae har en typisk Gram-positiv cellevegg og en ovoid morfologi. Celleveggsyntesen skjer som en kombinasjon av septal og perifer syntese i en såkalt tomaskineri-modell. Blant de mange komponentene som er assosiert med celleveggsyntesemaskineriet hos pneumokokker har de penicillinbindende proteinene (PBPene) sentrale roller. Klasse A PBPer (PBP1a, PBP1b og PBP2a) har både transglykosylase- og transpeptidaseaktivitet, mens de essensielle klasse B PEPene PBP2x og PBP2b kun har transpeptidaseaktivitet. Mutansjoner i PBPene har vist seg å føre til resistens mot penicillin og andre β-laktamantibiotika. Den økende spredningen av penicillinresistens fører til problemer i forbindelse med behandling av pneumokokksykdommer. PBP2b og PBP2x er essensielle proteiner hos pneumokokker, og studier har vist at delesjon av både PBP1a eller PBP2a er letalt for S. pneumoniae. I dette arbeidet har ComRSdepletionsystemet blitt brukt til å studere interaksjonene mellom PBP1a og PBP2a, og mellom klasse A PBPene og klasse B PBPene PBP2b og PBP2x. Vekst og morfologistudier har vist flere indikasjoner på at PBP1a har en sentral rolle blant klasse A PBPene. Fra litteraturen vet man at PBP2a og PBP1a i en viss grad kan vikariere for hverandre, men resultatene fra dette arbeidet tyder derimot på at PBP2a ikke er en fullverdig erstatter for PBP1a. Videre ble det vist at PBP1a har en nøkkelrolle i samarbeid med klasse B PBPene. Vekst- og morfologistudier indikerte at det er en interaksjon mellom PBP2x og PBP1a i den septale celleveggsyntesen. Studiene viste også at PBP1a og PBP1b trolig er lokalisert sammen med PBP2b i det perifere celleveggsyntesemaskineriet. Disse resultatene bidrar til å styrke tomaskineri-modellen da studien er gjennomført med andre metoder enn det som tidligere har blitt brukt. Abstract The main function of the bacterial cell wall is to withstand osmotic pressure and to maintain the bacterial shape and structure. Peptidoglycan is the scaffold of the cell wall and although its three-dimensional structure is thoroughly studied, the complete structure of this great macro molecule is still not known. Bacterial cell division and cell wall synthesis are tightly regulated and coordinated processes. These processes require precise localization of a variety of proteins. The Gram positive bacterium Streptococcus pneumoniae forms an ovoid shape by a two-state model with a combination of both peripheral and septal peptidoglycan synthesis. Several penicillin binding proteins (PBPs) participate in the biosynthesis of peptidoglycan. The class A PBPs (PBP1a, PBP2a and PBP1b) are bifunctional proteins that possess both transpeptidase and transglycoslase activities, whereas class B PBPs (PBP2b and PBP2x) are monofunctional transpeptidases. Changes in the PBPs are associated with β-lactam resistance. The dramatic increase in penicillin resistant S. pneumoniae causes challenges in the treatment of pneumococcal diseases. PBP2b and PBP2x are found to be essential proteins in S. pneumoniae. Several studies have shown that the simultaneous deletion of both PBP1a and PBP2a cannot be tolerated by the pneumococcus. Through experiments in this work the ComRS system was used to study the interactions between PBP1a and PBP2a. The system was also used to investigate the interactions between the class A and the class B PBPs. Growth experiments combined with studies of cell morphology indicates that PBP1a is the major class A PBP. From the literature we know that PBP1a and PBP2a can substitute for each other. The results from this work, however, showed that PBP2a cannot fully substitute all the functions that PBP1a perform. Results obtained in this work indicate that PBP1a interacts with PBP2x in the septal cell wall synthesis. The results also suggest that PBP1a, together with PBP1b, are co-located with PBP2b in the peripheral cell wall machinery. This work therefore supports the locations of PBP1a, PBP2b and PBP2x presented in the two-state model.no_NO
