dc.description.abstractEn metode for separering av enantiomerene til de kirale klorerte pesticidene α-HCH, o,p’ DDT, cis klordan, trans-klordan og oxy-klordan er blitt utviklet. GC-MS ble benyttet til kvantitativ og kvalitativ analyse av enantiomerselektiv akkumulering av pesticidene i filet av oppdrettslaks og torskelever hentet fra ulike områder i landet. Metodeutviklingen gikk ut på å undersøke separasjonen av kirale pesticidstandarder på ulike kirale kolonner og med ulike temperaturprogram og MS-program. Separasjon av enantiomerene ble oppnådd på en BGB172-kolonne med β-cyclodextrin-stasjonærfase og med masseanalyse ved m/z = 181, 235, 375 og 387. Med resultatene fra pesticidstandardene ble retensjonstid, retensjonsrekkefølge og separasjonsfaktor bestemt for enantiomerene. Det ble bestemt at (+) enantiomeren eluerer før (-)-enantiomeren for cis-klordan, trans-klordan og oxy-klordan på BGB172-kolonnen. Fiskeprøvene ble analysert på den utviklede metoden, og målanalyttene ble detektert og sammenlignet med standardene. Den enantiomerspesifikke akkumuleringen av hvert pesticid ble undersøkt ved å beregne enantiomerfraksjonen til pesticidene i hver prøve. For cis-klordan ble det oppnådd separasjon med god oppløsning og godt signal i forhold til støy. Det ble observert en tendens til racemisk akkumulering av cis-klordan, med EF-verdier nær 0,5, uavhengig av fisketype og sted. Det ble ikke detektert trans-klordan eller oxy-klordan i noen av prøvene. For akkumuleringen av α HCH var resultatene mer varierte, og ingen klare trender ble bestemt. Det kunne likevel observeres enantiomerselektiv akkumulering av pesticidet i både oppdrettslaks og torskelever, da EF-verdiene avvek fra 0,5. Den kromatografiske oppløsningen av α HCH var lav, og signalet var svært lavt i forhold til støyen, slik at resultatene inneholder en del usikkerhet. Det ble observert at akkumuleringen av o,p’-DDT også skjedde enantiomerselektivt i fiskene. En tendens til foretrukket nedbryting av (+)-o,p’-DDT, mens (-)-o,p’-DDT ble akkumulert kunne observeres, med EF-verdier under 0,5. For o,p’-DDT var det generelt god oppløsning, men i enkelte tilfeller var det vanskelig å bestemme enantiomertoppene, og retensjonstidene varierer fra standarden, så resultatene for o,p’-DDT er noe usikre. Det behøves å analysere flere fiskeprøver for å kunne bestemme klare trender av enantiomerselektiv akkumulering av de kirale pesticidene. MS-systemet som ble benyttet var heller ikke sensitivt nok for analysen i denne oppgaven.   A method for separation of the enantiomers of the chiral chlorinated pesticides α-HCH, o,p’ DDT, cis chlordane, trans-chlordane and oxy-chlordane has been developed. GC-MS was used for the quantitative and qualitative analysis of the enantiomer selective accumulation of the pesticides in farmed salmon and cod liver collected from different locations. The method development was pursued by studying the separation of the chiral pesticide standards on different chiral columns and with different temperature programs and MS programs. Separation of the enantiomers was accomplished on a BGB172 column with a β cyclodextrin stationary phase and with mass analysis of m/z = 181, 235, 375 and 387. With the results from the pesticide standards retention time, retention order and separation factor of the enantiomers were acquired. It was concluded that the (+)-enantiomer eluates quicker than the (-)-enantiomer in the case of cis-chlordane, trans-chlordane and oxy-chlordane on the BGB172 column. The fish samples were analyzed on the developed method and the analytes were detected and compared with the standards. The enantiomer specific accumulation of each pesticide was examined by calculation of the enantiomer fraction of the chiral pesticides in each sample. Separation of cis-chlordane was achieved with good resolution and high signal-to-noise ratio. There was a tendency of racemic accumulation of cis-chlordane, with EF values close to 0.5, regardless of the fish type and location. The results showed no detection of trans-chlordane or oxy-chlordane in any of the samples. The results from the analysis of α-HCH showed more variation regarding the accumulation, and no specific trends regarding fish type or location could be determined. EF values different from 0.5 was however an indicator of enantiomer selective accumulation in both farmed salmon and cod liver, with a majority of EF values below 0.5. A poor chromatographic resolution of α-HCH and a low signal-to-noise ratio has led to results of some uncertainty. There were also observations of enantiomer selective accumulation of o,p’-DDT in the fish samples. The results showed preferred depletion of (+)-o,p’-DDT, while (-)-o,p’-DDT was accumulated, with EF values under 0.5. The resolution of o,p’-DDT was generally good, but there were difficulties regarding the determination of some of the enantiomer’s maxima, and the retention times differed slightly from the standard. Hence there exist some uncertainties within the results. To determine specific trends of enantiomer selective accumulation of the chiral pesticides further analysis of more sample material is needed. The MS system did not perform with the sensitivity required for the analysis in this thesis.no_NO
